How to Make Liquid Culture for Mushrooms

Liquid culture (LC) is a nutrient-rich broth used to propagate mushroom mycelium. A small piece of tissue, agar culture, or spore syringe is added to the sterile broth, and the mycelium colonizes the liquid over 1-3 weeks. The resulting culture can then be used to inoculate grain spawn, substrate bags, or additional liquid culture jars.

Honey water is the most popular liquid culture recipe for home growers. Light honey (not raw or unfiltered) is dissolved in water at a ratio of 3-4% by weight. The solution is sterilized in a pressure cooker at 15 PSI for 20-30 minutes, then cooled and inoculated.

Dextrose (corn sugar) is preferred by commercial growers because it produces a clearer broth that makes contamination easier to detect. The standard concentration is 3-4% by weight.

Potato dextrose brothadds extra nutrients from potato starch, supporting faster mycelium growth for some species. It's commonly used for more demanding species like lion's mane and reishi.

For best results, use jars with self-healing injection ports and synthetic filter caps for gas exchange. This allows you to inoculate and draw culture without opening the jar, minimizing contamination risk.
